The world-famous gentleman thief Lupin III and his partner Jigen Daisuke successfully steal a massive sum from the vault of the Monte Carlo state casino and escape by car, shaking off their pursuers. Although the two are practically buried under piles of cash inside the vehicle, Lupin's keen eye detects that these are "Goethite" bills, the most exquisitely crafted counterfeit notes in history. Adhering to his principle of "never stealing fakes," Lupin carelessly scatters the Goethite bills on the road and sets his next target: uncovering the secret behind them. Thus, Lupin heads toward the mysterious European independent nation, the Grand Duchy of Cagliostro.
Lupin and Jigen change disguises to infiltrate the Grand Duchy of Cagliostro, but their car gets a flat tire along the way. While fixing the tire, they spot a girl in a wedding dress being chased by thugs. They intervene, drive off the attackers, and save the girl as she falls off a cliff. However, the girl, who left behind a ring engraved with Gothic script, is taken away by another group. It is later confirmed that she is Clarisse de Cagliostro, the heir to the grand ducal family of Cagliostro. Currently, due to the sudden death of the reigning Grand Duke, Count Luger de Cagliostro serves as regent, leaving the throne vacant. Lupin also recalls meeting young Clarisse once during his early days as a thief.
Luger, the de facto ruler of the Grand Duchy of Cagliostro, plans to marry Clarisse to seize the title of Grand Duke and establish a dictatorship. Once again imprisoned, Clarisse is confined within Luger's residence, the Castle of Cagliostro. To rescue her, Lupin calls upon his old comrade Ishikawa Goemon. Meanwhile, Inspector Zenigata, having learned that Lupin sent a calling card to Luger, leads his police squad (the Saitama Prefectural Police Mobile Unit) to arrest Lupin. Including Fujiko Mine, who has already infiltrated the castle disguised as a governess, the entire "Lupin Family" is now assembled. On the stage of the Castle of Cagliostro, a chaotic battle ensues to rescue Clarisse and unravel the mystery of the Goethite bills.
